Abstract
The Callipyge (CLPG) gene is associated with muscle development in sheep and may serve as a molecular marker for genetic characterization of meat-producing sheep. This study aimed to detect polymorphism in the CLPG gene in Cross Texel and Dorper F1 sheep using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). A total of 95 DNA samples, consisting of 4 Cross Texel and 51 Dorper F1 sheep, were analyzed. A 615 bp fragment of the CLPG gene was amplified by PCR and subsequently digested using the NaeI restriction enzyme. The digested products were separated by agarose gel electrophoresis to identify genotypes based on restriction patterns. NaeI digestion produced fragments of 400 and 215 bp. Two genotypes were identified based on the observed banding patterns. The GG genotype, represented by two fragments of 400 and 215 bp, was observed in 46 Dorper F1 and 39 Cross Texel samples, whereas the AG genotype, represented by three fragments of 615, 400, and 215 bp, was observed in 5 Dorper F1 and 4 Cross Texel samples. No AA genotype was detected. The results indicate CLPG polymorphism in both populations and demonstrate that PCR-RFLP using NaeI can characterize CLPG variation in these sheep populations.
