Antioxidant activity, Alpha-Glucosidase Inhibitory, Total Phenolic content and Total Flavonoid Content of Ethanol and Methanol Extracts of Aerva sangunolenta Sampled From Solo, Indonesia. 1) Faculty of Pharmacy Universitas Indonesia, Depok 16424, West Java, INDONESIA Abstract This study aimed to examine the impact of two different solvents, ethanol and methanol, in assessing the antioxidant activity, total phenolic content, total flavonoid content and alpha glucosidase inhibitory activity of Aerva Sanguinolenta samples collected from Solo, Indonesia. Antioxidant activity was measured using the 2,2 diphenyl 1 picrylhydrazyl assay method. The chemical composition of Aerva sanguinolenta was represented as total phenolic content and total flavonoid content. The antidiabetic activity of the ethanol and methanol extracts and the resulting fractions were examined by alpha glucosidase inhibition bioassay. All statistical analyses were performed using Graphpad prism 9 for Windows, Version 22. All data are presented as mean standard deviation and all data were analyzed using pearson regression model. The antioxidant activity of the ethanol extract of Aerva sanguinolenta showed inactivity with an IC50 value of 286,11, while the methanol extract had an IC50 value of 102,76l. In comparison, synthetic Vitamin C showed an IC50 value of 5.31 in the experiment.The results of this study showed the presence of substantial alpha glucosidase inhibitory activity for the ethanol extraction (14.8), while no alpha glucosidase inhibition was found in the methanol fraction.The total phenol content in the different extracts, expressed as gallic acid equivalents (GA) per gram of dry extract, ranged from 37.490 to 47.779 mg.
